Physico-chemical and Phytochemical Study & Microbial Analysis of Lemongrass Hydrosol

Cymbopogon citratus commonly known as lemon grass is claimed to possess diverse medicinal value among different cultures. The present study determined the phytochemicals and evaluated the antioxidant potential of Cymbopogon citratus leaves. The phytochemical analysis of the hydrosol was carried out using standard methods. The phytochemical analysis revealed the presence of flavonoids, tannin, saponion, terpenoids and other phenolics compounds.

Medicinal plants have great importance to the individual's health. The medicinal value of plants lies in their containment of some natural compounds, the most important of these bioactive constituents of plants include alkaloids, tannins, flavonoids, phenolic and others.

Lemongrass, (Cymbopogon citratus) an aromatic herb. The plant also contained flavonoids which were phenolic compounds that serve as flavouring ingredients of plant leaves, higher contents of tannins, flavonoids and terpenoids were observed than coumarin in lemongrass (C.citratus), Isoorientin, Caffeic acid, Quercetin and Chlorogenic acid. Lemongrass was found in folk remedy for coughs, consumption, malaria, pneumonia and vascular disorders. Researchers have found that lemongrass holds antibacterial, antifungal, antitumoral, and anticancer and insecticide activities and even the antimicrobial activity. The aim of study is to examine the phytochemical screening, and physico-chemical analysis of lemongrass hydrosol.

The present work evaluates the herbal waters/ hydrosol of Lemongrass obtained as innovative commercial products of a steam distillation process.

The following tables provide records on the most commonly tested physico-chemical, phytochemicals, and Microbiological evaluation in Lemongrass Hydrosol.

1.4 Result of Phytochemical Analysis of Lemongrass Hydrosol

The following result were obtained after performing the phytochemical test:

This method includes the selective and successive extraction of the plant phytochemical. The analysis of the presence of the main group of natural constituents present in the plant extract was done using different specific reagents. Chemical tests were done to identify bioactive compounds of pharmacological importance through standard methods. Phytochemicals such as mentioned below were qualitatively determined.

S.No. Phytochemical Tests Lemongrass Hydrosol Specification Results
1. Flavonoids Highly presence of red colour (+ve)
2 Tannin Highly presence of green colour (+ve)
3 Saponin Presence of foam formation (+ve)
4 Terpenoids Highly presenmc of reddish brown colour (+ve)
5 Coumarin No presence of yellow colour (-ve)
6 Steroids Highly presence of yellowish/greenish fluorescence (+ve)

1.5 Result of Microbial Analysis of Lemongrass Hydrosol

Microbial Specification Results
Aerobic Mesophilic Bacterial Count <100 CFU/g Conforms
Yeast and Mould <10 CFU/g Conforms
  1. Appearance- The Lemongrass hydrosol/aromatic water is in a liquid form similar to water. That is because it is soluble in water.
  2. Colour- The Lemongrass hydrosol is a clean, colourless to slightly cloudy liquid.
  3. Odour- The odour of a lemongrass hydrosol is derived from the leaves of a lemongrass plant. The odour of Lemongrass herb water has a fresh, grassy, lemony mildly herbal aroma. However the aroma changes from season to season with variety in its volatile compounds due to various factors like - temperature, rainfall, humidity and soil health etc.
  4. Solubility- It is soluble in water and alcohol but insoluble in fat/fixed oils.
  5. pH- A pH of 7 is neutral, above 7 the pH is alkaline and below 7 the pH is acidic.
  6. Acidity/acidic Lemongrass floral water is acidic in nature as it has a low pH, as opposed to a base, which has a higher pH.
  7. Specific Gravity is used to define the weight of a liquid as compared to density at a specific temperature. If the specific gravity for a Lemongrass hydrosol is less than 1, that means the material will flow in the reference substances, and if the water sample is more than 1, that means the material will sink in the reference material.
  8. Heavy Metals this test is done to check for irregular levels of toxic or potentially harmful metals. The Heavy Metal Limit test is based on the reaction of metallic impurities with hydrogen sulphide or sodium sulphide in an acidic medium to provide metal sulphide which provides brown coloration.
  9. Microbial Test is important and required by many industries worldwide, where human health is at risk of being adversely affected by disease-causing bacterial & virus, and microbial toxins.

Microbiological evaluation of a sample is the use of biological, biochemical, molecular or chemical methods for the identification of microorganisms in a sample.

Observations of Phytochemical Tests for Lemongrass Hydrosol

1. Flavonoids

Formation of red colour indicates the presence of flavonoids.

2. Tannin

Formation of green colour indicates the presence of tannin.

3. Saponion

Formation of foam indicates the presence of saponin.

4. Terpenoids

Formation of reddish-brown colour indicates the presence of terpenoids.

5. Coumarin

There is no presence of yellow colour which shows there is no presence of coumarin.

6. Steroids

Formation of yellow-greenish fluorescence indicates the presence of steroids.

The study revealed the presence of several phytochemicals present in lemongrass hydrosol like flavonoids, tannins, terpenoids and phenolic compounds constituents which play a role in human health and wellness.
